Inhibition of Tumor Cell Glycolysis

چکیده

The discovery of the active group of the hydrogen-transferring enzymes — nicotinamide — for which O tto W a r b u r g was nominated by the Nobel Committee 1 “for a prize for the second time in 1944, when he again was found to deserve the honor”, has had a profound influence on all of biochemistry. The present paper describes a fundamental inhibition of intracellular tumor meta bolism produced by reduced diphosphopyridine nucleotide (DPNH2) added extracellularly.

Methionine-sensitive glycolysis in transformed cells.

Glycolysis in several tumor cell lines grown in tissue culture was inhibited by methionine. Kirsten murine sarcoma virus-transformed rat kidney cells (K-NRK) were inhibited 60-75% by 10 mM methionine, whereas normal rat kidney (NRK-49F) cells showed little or no inhibition. Inhibition of glycolysis in K-NRK cells was manifest 2-4 hr after exposure to the amino acid. Glycolysis in a chemically t...

Exploiting metabolic inhibition to eradicate residual tumors after chemo-immunotherapy

Cancer cells have long been known to exhibit metabolic reprogramming which involves a shift towards glycolysis. The enhanced glycolysis of cancer cells not only impose nutrient restriction in the tumor microenvironment, but also contribute to the acidic environment which is hostile to the invading anti-tumor immune cells. Hence, targeting the glycolysis dependency of tumor cells has been exploi...

Under normoxia, 2-deoxy-D-glucose elicits cell death in select tumor types not by inhibition of glycolysis but by interfering with N-linked glycosylation.

In tumor cells growing under hypoxia, inhibiting glycolysis with 2-deoxy-d-glucose (2-DG) leads to cell death, whereas under normoxic conditions cells similarly treated survive. Surprisingly, here we find that 2-DG is toxic in select tumor cell lines growing under normal oxygen tension. In contrast, a more potent glycolytic inhibitor, 2-fluorodeoxy-d-glucose, shows little or no toxicity in thes...

Glycolysis is the primary bioenergetic pathway for cell motility and cytoskeletal remodeling in human prostate and breast cancer cells

The ability of a cancer cell to detach from the primary tumor and move to distant sites is fundamental to a lethal cancer phenotype. Metabolic transformations are associated with highly motile aggressive cellular phenotypes in tumor progression. Here, we report that cancer cell motility requires increased utilization of the glycolytic pathway. Mesenchymal cancer cells exhibited higher aerobic g...

The miR-383-LDHA axis regulates cell proliferation, invasion and glycolysis in hepatocellular cancer

Objective(s): To explore the correlation between expression patterns and functions of miR-383 and LDHA in hepatocellular cancer (HCC). Materials and Methods: We detected the expression of miR-383 and LDHA in 30 HCC tissues and their matched adjacent normal tissues using qRT-PCR. Then we performed MTT assay, foci formation assay, transwell migration assay, glucose uptake assay and lactate produc...

The role of glycolysis in the growth of tumor cells. II. The effect of oxamic acid on the growth of HeLa cells in tissue culture.

The finding that potassium oxamate inhibits glycolysis by ascites tumor cells in vitro, without inhibiting oxidative metabolism (I), indicated that oxamate might be applied to the study of the role of glycolysis in tumor growth in viva. If, as Warburg (2, 3) states, tumor cells derive much of their energy for survival and growth from glycolysis, then the inhibition of glycolysis in tumor-bearin...
